A Nemesis is an NPC you create who will be involved in your champion's career beginning from level 25.
Creating your Nemesis
At level 25 you will receive an email from MCPD Captain Martin that directs you to the MCPD Headquarters (6920 246 2182). MCPD Headquarters is one full block north of City Hall in Millennium City, follow the east side of the building to the north.
Notes
Just want to add that quests are designated by your level. It is possible to be a level off if you get the item, then level and activate it for the quest. I don't think there's any set order for the quests, but there may be a set count of the random quests before the actual story ones kick in. It may also be possible that some of the indented missions can be activated from other missions if you happen to get it at that point, this is more for reference.
