There are number of missions in Champions Online that are granted from an item in your inventory. These items will have notation when you hover over them that reads "This item starts a mission".

In order to obtain the mission, you most double-click the item and then accept the mission.

Items are obtained by drops from random faction members, interacting with certain objects in specific locations (also listed as Location Missions) or as rewards for defeating a specific villain.
